Abstract:
This project aimed to examine if the novel sampling technique of a Whatman® FTA®
card can provide DNA profiling results rapidly from body fluids on substrates that are
difficult to obtain DNA profiling results from. This project also aimed to determine if a
soaking process could obtain more DNA and better DNA profiling results from unfired
and fired cartridges.
Examples of forensic samples are body fluids; and touched samples containing trace
level amounts of DNA. It is important that an optimal sampling technique is used that
can increase the amount of DNA recovered from the substrate or improve the quality
of the DNA profile by increasing the number of alleles or by increasing allele peak
heights. The sampling technique combined with an optimal PCR process, standard PCR
or direct PCR workflow, is also examined in this research......
