Abstract:
The Bcl-2 overexpression is a hallmark of follicular lymphoma (FL). Since patients with FL often suffer from progressively resistant to chemotherapy refractory disease, the development of new regiments is required. In this study we analyzed for the first time the effects of a Bcl-2 antagonist, HA14-1, alone and in combination with antineoplastic agents commonly used against follicular lymphoma, in human FL cell lines with t(14;18). The cell lines were established from patient samples send for PAD analysis and earlier analysed in detail. All cells were sensitive to HA14-1 induced cytotoxicity (the LC50s of 4.5 to 12.6μM) and apoptosis. Moreover, HA14-1 enhanced dexamethasone and doxorubicin mediated (in schedule independent and dependent manner, respectively) cytotoxicity and apoptosis, but antagonized with vincristine, in all FL cells. Together these findings warrant further in vivo studies and provide conceptual basis for therapeutic intervention in follicular lymphoma.
